Abstract

The reactions of metal saphen complexes with plasmid pUC 19 were observed under aerobic reducing, oxidative, and oxidative-reducing conditions using agarose gel electrophoresis. Plasmid cleavage was only observed under oxidative conditions with peroxide present, but greatly accelerated when the reducing agent ascorbate was also added. It appears a catalytic cycle occurs when both ascorbate and peroxide are present, since the cleavage is observed at only 200 nM saphen complex. Rate constants for the degradation of supercoiled plasmid were calculated, and first-order kinetics were observed with respect to concentration of metal complex and plasmid. This suggests a bimolecular process in which the complex and DNA must enter close proximity, possibly through the intercalation of the ligand into the DNA strand. The iron and copper complexes likely have higher rate constants.
